Cryoprotection of porcine cornea: a scanning electron microscopy study
نویسنده
چکیده
Porcine corneas were frozen with Me2SO, glycerol, 1,2-propanediol and PEG-400. The effects of the range of concentrations (5% and 10%) and temperature regimen (1oC/min and 5oC/min) were investigated. The integrity of corneal endothelial cells was evaluated by scanning electron microscopy and trypan blue staining. The presence of 5–10% PEG-400 in the protective medium was the most effective in minimizing changes in the integrity of the corneal endothelium during freezing-thawing procedures.

Human corneal equivalent as cell culture model for in vitro drug permeation studies.
AIMS For the study of transcorneal in vitro permeation of ophthalmic drugs, excised animal cornea or corneal epithelial cell culture are frequently used as a replacement for the human cornea. The main purposes of this study were to reconstruct a complete human organotypic cornea equivalent, consisting of all three different cell types (epithelial, stromal, and endothelial); to test the barrier ...

متن کاملWavelength-dependent conformational changes in collagen after mid-infrared laser ablation of cornea.
We ablated porcine corneas with a free electron laser tuned to either 2.77 or 6.45 microm, two matched wavelengths that predominantly target water and protein, respectively. The ejected nonvolatile debris and the crater left behind were examined by circular dichroism, Raman spectroscopy, and scanning electron microscopy to characterize the postablation conformation of collagen proteins. We foun...
متن کاملThe Effect of Lyophilization on Light Transmission of Amniotic Membrane: A Comparison with Rabbit Cornea
Background & Aims: Amniotic membrane persists for a long time after ocular transplantation (as corneal substitute) and can affect light transmission (transparency). The aim of this study was to evaluate the transparency of amniotic membrane after freeze-drying (lyophilization) and to compare the results with transparency of rabbit cornea. Methods: Transparency of rabbits’ corneas and fresh and ...
متن کاملDevelopment of a rabbit corneal equivalent using an acellular corneal matrix of a porcine substrate
PURPOSE The tissue equivalent that mimics the structure and function of normal tissue is a major bioengineering challenge. Tissue engineered replacement of diseased or damaged tissue has become a reality for some types of tissue such as skin and cartilage. The tissue engineered corneal epithelium, stroma, and endothelium scaffold are promising concepts in overcoming the current limitations of a...
